Makidotvn/iStock via Getty Images Kodiak Sciences ( KOD ) added ~33% in the premarket on Thursday after the company announced that a Phase 3 trial for its eye disorder therapy, Zenkuda, reached key goals in patients with diabetic retinopathy, an eye complication of diabetes. Citing 48-week data, the Palo Alto, California-based biotech said Zenkuda reached its GLOW2 trial’s primary endpoint as nearly 63% of trial subjects experienced a ≥2-step improvement in the diabetic retinopathy severity scale (DRSS) with statistical significance. In comparison, about 3% of those in the sham arm of the study experienced ≥2-step improvement in DRSS. The anti-vascular endothelial growth factor (VEGF) therapy also demonstrated statistically significant superiority to sham in terms of several secondary endpoints, including one related to reducing the risk of developing prespecified sight-threatening complications. Safety data indicate that the Zenkuda intravitreal injection was well tolerated among patients with a low occurrence of common ocular adverse events. Building on the success of the company’s pivotal GLOW1 study for Zenkuda in DR, Kodiak ( KOD ) had expanded GLOW2 to include those with proliferative diabetic retinopathy and mild diabetic macular edema in addition to patients with moderately severe to severe DR. Patients in both studies received either sham injections or Zenkuda 5 mg with an ultimate treatment interval of every six months. Given positive GLOW2 results, Kodiak ( KOD ) said it has “a multi-indication Biologics License Application (BLA)-ready profile," and it intends to speed up BLA submission plans. ( KPTI ) for quite a while now with optimism that has not matched reality, placing “Buy” sentiments on the company in 2024 and then later that same year , only to see the valuation of the stock decline by more than 50% as the market has awaited the results of a key, arguably existential, phase 3 trial. We now take a first look at the results of that trial, and the market has not been pleased, sending the share price from a high of $8.90 to the near-$5 price point that it is at the time of writing. Today, I want to take another look at the company, intending to outline why I think there is reason for optimism here. Pipeline Updates Selinexor For years now, KPTI has been working ardently toward finding new niches for its XPO1 inhibitor, selinexor outside the multiple myeloma space. A readout for endometrial cancer remains a bit on the horizon, with the latest guidance from management suggesting that results should be coming in mid-2026. This relates to the phase 3 XPORT-EC-042 trial evaluating selinexor as a maintenance therapy for TP53 wild-type endometrial cancer; recall that this trial builds on a signal from the SIENDO study, which met its PFS endpoint but was not considered clinically relevant, but a signal of major benefit was identified in the TP53 wild-type subgroup of patients. So we will wait for the results of XPORT-EC-042 with bated breath. The news we have today relates to a topline readout of the phase 3 SENTRY trial, which compared the addition of selinexor to ruxolitinib versus ruxolitinib alone for patients with treatment-naive myelofibrosis. The trial met only one of its two primary endpoints. Adding selinexor improved the rate of patients achieving a 35% reduction in spleen size (rates of 50% and 28% for combination and ruxolitinib alone, respectively), but the co-primary endpoint of total symptom score was not improved (9.89 vs. 10.86 point i...

The European Union opened an investigation into Snap Inc. ’s social network and escalated a probe into a number of pornography platforms as the bloc ramps up efforts to protect children online. The Snapchat inquiry will look at how the platform verifies its users’ age, how it tackles grooming and criminal activities that target underage users, and how it removes content promoting drugs and other illegal items, the EU’s executive branch said in a statement Thursday. In parallel, the European Commission said it will move forward with a previously announced investigation into pornography platforms PornHub , Stripchat, XNXX and XVideos. The second stage of the probe comes after the commission preliminarily found the companies broke the law for not effectively preventing minors from accessing their websites. The EU’s moves come as social media companies and other internet platforms are facing increasing pressure for allegedly harmful and addictive design around the world. In a landmark case in the US, Meta Platforms Inc. and Alphabet Inc. were ordered by a jury to pay damages to a 20-year-old woman who claimed her addiction to social media caused her mental health struggles. The verdict in the US will “give a very clear message that online platforms have to take seriously the risks that they are posing,” the commission’s top technology official, Executive Vice President Henna Virkkunen, said at a briefing about the investigations. The commission’s probes were brought forward under the Digital Services Act, or DSA, which is the EU’s content moderation rulebook binding online platforms to standards of risk-mitigation and transparency. Companies under investigation can offer commitments to address the commission’s grievances, or potentially face fines of as much as 6% of their annual global sales.
